Tohru and Yuujirou are from Princess Princess (manga about crossdressers. really really fun~!). Soushi and Kazuki are from Soukyuu no Fafner (or Fafner in the Azure).

*squees* Just watched all of the anime, and what I could find of the manga of Princess Princess - for me, it was easier to say that Tohru looked like a girl when not in costume in the anime, than the manga. And Akira's eyes freaked me out.

Is there a lot of difference between the anime and manga?

just a little. the manga goes a little further than the anime did (anime stopped at 'Tohru's changing schools!' while that happened only in volume 3)
I'm not sure on the anime, but the manga was supposed to be a BL story, but the publishers didn't want that (...publishers were stupid; there'd been another BL story before), so there are a few hints...I think more-so than the anime.
