Demons have long been integral to the SPN arc, as foes and uneasy allies, and the striking black - or yellow, or red, or white - possessed eyes are a striking indicator of these creatures. And this striking effect has been used very well on familiar characters throughout the series - it's so jarring and fascinating to see a familiar character

I was looking at another fic after I saw this and one crossed my mind, although at that time we'd only known her for that episode - what about Tessa being possessed by Azazel in In my time of Dying (2.01)?
