So, there was the comment that we're a well-behaved community due to the lack of shipping wars given that there's really only one viable pairing in the movie to begin with. I agree wholeheartedly. Minion would sacrifice himself to save Megamind or Roxanne because he couldn't live without his Master or with him grieving and heartbroken.

Honestly? I think Megamind would have an easier time of dealing with his choice if he had chosen to save Minion. I can't really articulate why, but...

I guess it would all depend on how far along in the relationship, if there was a relationship, Megamind and Roxanne were. Pre-movie? Minion. Immediately post-movie? Minion. Late in the game? Welllll, that's a harder one to answer. Roxanne. I think.

My full and honest answer is that I think he would try to save both of them, and die in the attempt.

On the whole, I think he'd refuse to choose unless there was absolutely no other option and it was the last second. It's part of his never giving up even in the face of certain defeat personality. If in that last second he had to choose, I believe it would be Roxanne, because Minion is his bro, and bros go down fighting together. Roxanne, kick ass though she may be, is still a lady, and both Megs and Minion are strange gentlemen, in their own ways. Minion would probably be trying to save the damsel, too, especially if there was no threat to Megs himself, and I believe Minion would expect Megs to save her. It's the polite thing to do, women and children first. Weirdly old fashioned, but Minion is certainly an old fashioned fish. Just look at his apron and wig and his little curtsey as Space Step Mom. XD

Both Minion and Roxanne would tell Megs to save the other, since they're both selfless characters. But I just have a feeling Megs would save Minion. Yes, Megamind loves Roxanne, but he's been with Minion his whole life, they're practically brothers. He would probably defuse the bomb on Minion first, and tell him to get Roxanne to safety while he deals with (Insert Villain of choice here).

Hypothesis: if death was 100% certain, he'd save Minion, but if there was the slightest chance of the person he didn't save being able to save themself, he'd save Roxanne because he knows Minion has greater self-saving capabilities. (He ought to know; he built them into that suit.)

That's exactly what I was thinking, too. It really depends on the particular life-threatening situation - not to put down Roxanne, but most of the time, Minion is better equipped to save himself.
