Wow, there is a ridiculous amount of awesome fic in both TOS and Reboot fandom. I could probably write a two- or three-page list of K/S stories that I'd like to see podficced, but if I have to pick just one for the sake of argument? I'd say Breaking Points, by ragdoll987. It's long, plotty and generally amazing - one of my very favorite stories in this fandom. =)
Like _ryouseiteki_ said, Observations by anon_j_anon. It is the best piece of fanfiction I've ever read, of any fandom, but it is ridiculously long. And lots of it would be very difficult to read because it does into stream-of-consciousness mode, broken phrases and run-ons and just very very emotional, beautifully written pieces that could never be done right. Plus, almost all of it is written in 1st person Spock POV so the Spock voice would need to be PERFECT.
And then it's so heart-wrenching during certain chapters that I can't imagine anyone being able to read that aloud without breaking down right then and there. I'm proof-reading for it at the moment and I have to take continuous breaks because I can't take reading it all at once like that.
So yeah, even though Observations would be a dream come true, I almost don't want it done.
So that's not really an answer. I'll be back with a real answer, I swear!
Yeah, I'd love an Observation podfic also, but I could see that it would be really difficult to do, not simply because of its length.
That's another interesting point of discussion though: can someone ruin a podfic with their interpretation of it? I haven't run into a podfic I didn't like (if I liked reading the fic, I liked listening to it), but I suppose that the reader's way of doing things and a complixity of a story might make a story almost impossible to be done. Hmmm...interesting...
Actually, the first and only podfic I listened to, I hated. I won't say which it was, but the style of the story is sort of... colloquial and almost like a conversation, but the person sounded so mechanical and concerned with pauses and articulation that it just lost all flavor of the story and it just sounded extremely awkward. So I've never heard another podcast. I'd love a recommendation if you have any.

Yes!!! Thank you so much for pointing this out. Usually I'm a clear cut slash kind of girl , but this gen story is awesome!!! Yah!! This'll definately keep me busy for my next trip!
