Zed is a total nerd from the world as we know it. Up to 2009, life was normal until the zombie apocalypse happened. This also means, if you are a fictional character, he may, or may not know of you. Fourth wall all you want for Tim! If Zed's a fan of Batman comics he can say/think whatever about Tim, as long as he keeps in mind that this version of Tim is only 13 and would be seriously traumatized to know that his parents are going to die in a couple years, etc.

Zed is a passing fan of Batman. He spent more time with his head buried in Popular Science than he did comics, but he might recognize the name. He's not about to traumatize some 13 year old, he's pretty sensitive about that. There's enough trauma in the world, he really doesn't want to add to it.

Internal fourthwalling/recognition is fine for Cain if Zed was aware of BSG, although I highly advise against actually telling her she'd fictional. She wouldn't take it well :3

4th wall at will for John! Until Zed mentions the whole "oh yeah you're fictional" thing, John will probably assume he's just familiar with a version of him from some other timeline. One that apparently had zombies, but eh it's not completely improbable.

Zed can 4th wall with Dean too, if they ever meet, but don't expect Dean to take it well. We've seen how he reacts to that sort of thing in canon (There were 'fictional' books written about his life that he didn't know about until later and he is NOT happy about it) so finding out that his life is actually a TV show probably wouldn't go over like sunshine and roses. Zed could admit to having heard of him though, and as long as Dean can reasonably assume that it's because Zed knows some hunters or something, it'll be alright.
