So much of this show is the hilarious slips of the tongue, double entendre, or just plain snark-ass that the characters toss at one another. Not surprisingly, most of my favorites are from Giles and Spike. Since I don't remember half of the brilliant ones until I rewatch episodes, I'll just pick the one that sticks with me at all times.

Mister, if I wasn't in love with you before I most assuredly was after that. And I was in love with you from close to the first second after your first entrance. Librarian? Supernatural mystical awesome amazing librarian? Doesn't get much better. Ever.
The blueberry scone bit ranks up there too. And--if we're counting all the Buffyverse as the meme suggests--Spike's "fluffy puppy with bad teeth" speech from Angel is high on the list as well.
