PCR (polymerase chain reaction) why are you such a fickle mistress? One day you and I are working together so well, things look great, you have beautiful florescent bands that are caused by carcinogenic EtBr (it's alright I forgive you). But today after all the same steps we seem to not be in the mood to fluoresce. Have I done something wrong? Did I miss a step? Did I screw up the DNA extraction? Still you remain silent. Were the annealing temperatures not quite right? Didn't I give you the last eight hours of my life? But no, I will never know what I did wrong. All I can do is get up tomorrow try try again and hope that maybe you are in a better mood. PCR you truly are a fickle mistress.

~Well, at least the meet the new Dean of the Ag College Ice Cream Social went well. And chocolate syrup and rainbow sprinkles make the world a better place.~

(I know not terribly insightful but not all days can be brilliant streams of thought).
