To answer your question, no. Lestrade met Mycroft before he met Sherlock, so there wouldn't be a change in behavior to notice. The two have never interacted together in front of Sherlock. Mycroft gave Lestrade similar training to that which Sherlock gave John. Also, sherlock is a clueless idiot when it comes to emotions.
