1) Dean was so overprotective of Sammy all through their schooling years. Like, it worried teachers and kids just how self-sacrificing he was when it came to Sam. and he totally threatened the lives of any little kid that attempted to bully Sam ;P
2) Dean knew about Sam going to Stanford before Sam told them. and even though he was heartbroken that Sam was leaving him, he was damn proud of his baby brother, and happy that he was finally getting to live the normal life he wanted. and Dean also totally went to Palo Alto like, once a month to check up on Sam, just in case.
3) Even though it totally and completly fucked up Sam and their dynamic as a team, as brothers, Dean would never regret bringing Sam back to life, making the deal, and going to hell. And even though Ruby fucked him over 10ways to Sunday, I think Dean's sorta greatful to her in a way for keeping Sam alive.

alrighty.
1) I believe that he had his powers his whole life, that they didn't just start when he was 22. but they were dormant, only coming out in spurts when he was over emotional (like if he was really pissed lights would flicker or something would break..or something like that)
2) He really liked hunting when he was a kid. he liked being geek boy and researching and having John be proud of him. but he hated watching his family risk their lives all the time, so he wanted out.
3) Even though Dean said they didn't talk in 2yrs while he was in college, I totally think Sam kept tabs on his brother. tracked him down week after week to make sure he was ok, and alive...and he totally called Bobby every once in a while to double check, but swore the man to secrecy ;D

1) she wasn't always evil and the devil's bitch, that's just Kripke's way of covering up plot holes. She really wanted to help the boys at some point (some point being S3 and the first half of S4).
2) S3 Ruby totally wanted Dean.
3) She did love Sam or something kinda like love, but then Lilith totally mindfucked her by saying that if she made Sam go darkside and free Luci, she's be queen bitch or something.

aaaaand ok
1) in ITB, Mary knew Dean was her son...or was atleast somehow connected to him. I mean, I honestly can't imagine her telling a stranger that she doesn't want that life for her children, or agreeing to his promise. so, on some level atleast, she knew.
2) If Mary were still alive...she would've taught the boys to at least protect themselves. even if they weren't hunters, they'd probably have amulets and salt and at the very least, know to say 'Christo' and that static and cold spots= baaaad.
3) Mary knew about the angels...why else would she, being an (ex)hunter, say 'angels are watching over you'?
