With Photoshop and a pressure sensetive tablet. :) Setting the opacity of the brushes to be sensitive to the pressure applied on the tablet lets color be put on in increments instead of completely covering the colors beneath all at once, and gives nice textures while you're doing it. If you use a mouse you can get a similar effect by turning the opacity way down on the brushes and going over the same area with multiple strokes.
