Yup, you can die from lack of sleep. Sleep deprivation lowers white blood cell count, and I think other physical functions are also impaired. The record for the longest time spent awake (for a human) is eleven days. Source, although I originally heard this on House MD.

I think the longest I've ever been awake was around 36 hours; I can't imagine staying awake for 11 days.
