There's a scene in 1.22 'Devil's Trap' where Dean shoots a demon in the street. It's the colt, I don't know if that matters, but it is smoking. Dean is in the background blurry then the focus changes to him from the gun. I'm not sure of the timestamp but it's after they save John from the apartment where the demons are holding him.
