I think there is a scene like that in season three's Long Distance Call. Sam is tied to a chair by the MOTW, who is getting reading to stab him with a big knife. The MOTW lifts the knife high and there is a specific shot of the knife going down.

Oh and in Bloodlust from season 2, Gordon uses a big knife while torturing the female vampire leader, if I remember correctly there should some good knife shots, and Dean stabs a female demon quite frenzy like in the climatic scene in Malleus Maleficarum.

Also have a look at the season 5 opener where Bobby stabs himself and not Dean, there is a shot of just the knife there.
