I used to love and watch venture brothers, but I can't remember the story line at the end of season 1 and the beginning of season 2.

What happened that made doc venture go crazy?

The boys got killed.

Yeah, I went on wikipedia and quickly remembered that that was where we learned about the boys being death prone, and they're cloned on a regular basis.
