It's a sequel but you don't really need to have seen the first one, you just need to get the basic concept.

Idk anything about DC comics and I rarely watch DC movies so I'm prob wrong but my impression from the trailer was that the squad is a rotational lineup based on whatever villains they have wanting to be released from villain jail.
