Yes, if the writer claiming the prompt second is claiming it as their second or third prompt.
In order to maximize use of prompts, but also let people use whatever prompts inspire them, if you want to use an already claimed prompt you have to pick at least one other prompt to do as well.
Also, do robot versions of a character played by a different actress (but still the same person as a character) count for prompts. If it helps, I'm talking about Elizabeth Weir from Stargate Atlantis, after she got turned into a replictor and her actress was replaced.
You can pair your over-40 woman with anyone you like. The story just has to be *about* her, not about the person she's paired with.
As for the robot thing... woo. I don't know a lot about Stargate. Do replicators carry all the memories and self-identity perception of the original? If the replicator thinks of herself as Elizabeth Weir and has Weir's memories, then yes, she counts. Prompts are closed, though, but if the replicator Weir is Weir, you could take an existing Weir prompt and write about the replicator.

I had forgotten to join the community so I could post the stories here - er, if we are meant to post here?. I'll go ahead and post in my own LJ, and then cross-post once permission is granted.
I tried to tag the story I just posted, but the only tags visible to add are "definitions," "modpost," and "rules," and when I tried to add my own tags for author, characters, and fandom, they didn't show up. Can you check the community's tag permissions settings?
