I do a fairly similar job of patching the tubes on my bicycle. I hadn't thought of clamping them that way, though. I use vulcanizing compound like this: https://www.amazon.com/Rema-8oz-Patch-Glue/dp/B0017RYP4C which partially dissolves the rubber, and press the patch on with hand pressure, and by the time I get it back in the tire it's ready to hold air pressure.
Man, that's a complex rear wheel interface. Does it have a drum brake on one side, and the chain cog on the other?
I think your compound is far more appropriate for the job. I just didn't have one. So I just applied CA glue. And grinded a fair amount of rubber off of the tube and the patch. Which is also a piece of old tube.
Yeah, the wheel taking off process is slow, but putting back even longer. Every time you then have to tighten the chain and the drum brakes sping.
Yeah, I've been into drum brakes enough to see why everyone jumped on the disc brake trend. They're fussy. I use a bit of sandpaper for roughing up the tire and patch: I'm afraid of removing too much material. Then again, your tubes are probably 1.5mm thick, where mine are maybe one third of that.
