For the Master, you can look up a bit to the Doctor's loss of virginity.

River? She became Serenity's pilot and, of course, a thief along with the rest of the crew. One day, a job went a bit wrong, largely because the bit of "alien technology" they were stealing really was a bit of, well, alien technology. She ended up separated from the rest of the crew, accompanied by a handsome Time Agent who was trying to get the thing as well. They had to lie low until morning, when the Time Agent said he could get them out of there. She very sensibly suggested they have sex to pass the time. He agreed that would be a great idea and wasn't put off at all by the fact she was a virgin; he just said that meant he had to be extra good so she wouldn't settle for less in the future. She never did.
