To be honest, I'm not familiar enough with what they should look like to know the answer to your question, but I'm a big believer in "When in doubt, see the vet." At the very least, call the vet, describe the issue and ask whether they think you should bring him in.
Unless you've got a great hamster-vet, I'd monitor it every day and see if it's getting bigger. I've never seen that before, but I know little guys can be prone to tumors...
