Everyone else is doing this, so I will, too. If you don't mention a particular fandom, I'll assume that you're asking this as a general question

That's easy. Besieging Omelas, my Dresden Files crossover novel. It's the longest things I've written to date, and I think that it's the story that sounds the most like Butcher--genslash aside. (Anyone reading it for the romance or the sex would be disappointed, because very little actually happens. There's a little tease-flirting from John Constantine, which Harry ignores; two male-male kisses, one of which gets complained about; the most awkward declaration scene ever penned; and a sex scene that's more or less skimmed over. It's 99% action-adventure and trying to save innocents (and one not-so-innocent) from Hell--which I, frankly, loved writing.
If we're talking Shakespeare, it's a tie between Rosemary for Remembrance (which I summarized as "Hamlet in space, starring sensible and heroic Ophelia, genderswapped Claudius, clones Rosencrantz and Guilderstern and alien Fortinbras") and the clockpunk Gimmors and Devices, which I still want to turn into an actual book and which I'm rather afraid of messing with (though the middle and ending need to be developed a LOT more, I know).
Rosemary stems directly from my early love of space opera and science fiction mysteries. It's old-fashioned pulp, and I make no apologies for it because I rather love that. Also, even if the recipient hadn't asked for Heroic Ophelia, I would have written about her--most likely both her and Genderswapped Claudia, because I wanted them to have a chance to tell their own story. Plus, I like active heroines who actually affect the plot by their actions.
Gimmors--you know, I never even considered opening the story with anyone but Eleanor de Bohun, and I don't know why. From the minute that I got the idea--

I am grinning like crazy here. Not only did you get Vimes and Granny *and* Death all into the same story, you also had Vetinari and a Vimes-Granny confrontation and lots of Young Sam, and CATS -- either you're on my flist or you're just psychic, because this story was basically crammed full of all of my bulletproof Discworld kinks. And not only that, but I kept forgetting you're not Pterry -- you have the style down *perfectly*. It's like having a Discworld novella of my own!
So, yeah, thank you again. Best. Yuletide. ever.
