It's more like the way he worded it, "I've come to understand this is my life. And I love it." people realize that they are happy, but they don't come to understand that they love their lives? who is he trying to tell the story to? Charlie? himself?
Afterschool Special comes to mind, when the teacher asked Sam a simple question: are you happy? he couldn't come up with an answer. Speaking of Afterschool Special, "I am not evil, I'm not" that line, the little break in his voice, makes me so sad every time I think about it.
It's worth pointing out that, for the first time in a while, Sam has a lot of good things in his corner: a safe space in the Bunker, Dean's sudden affection, an unambiguous mission that's suited to his strengths, etc.
