Sam commits to “saving people, hunting things” with Dean as his true calling.

It seems to me that it was in this episode that the plot twist of the series took place. It began as "two brothers are looking for a father," and after this episode it becomes clear that "two brothers are hunting evil spirits," which as a result leads them to "Swan Song" and tragedy. Was Dean's last thought then that this choice was wrong? Why did he pull Sam out of university? I can't even imagine what Dean thought about all this at the time.

He lampoons being overcome with emotion at Sam’s speech to cover the fact that . . . he is overcome with emotion at Sam’s speech.

This is one of the best conversations the brothers have at the end of the episode.

Yes, absolutely. This episode was pivotal. It's here and in the next episode that we see the full yin/yang reversal take place. Sam ends this episode by committing to hunting while Dean begins the next one by literally and figuratively losing heart. By the end of Faith we are seeing signs that Dean is losing faith in his father and his father's mission. From then on it is Sam rather than Dean who drives the hunting.
Yes, I think there are signs in the latter part of the season that Dean starts to regret pulling Sam away from Uni, though he still wants his company of course. The pendulum swings back and forth between the brothers for the next 5 seasons but, yes, at the end of season 5 I think Dean is thoroughly disillusioned with the hero myth and more than ready to walk away from hunting.
I agree the final brother scene here is wonderful. In fact, I think the whole episode is one of the overlooked greats of Supernatural.

I've just been taking another look at the Bad Company lyrics and while it obviously plays over the last scene because Meg is bad company, I believe the full lyrics are doing double service in referring to the brothers' situation to and may foreshadow Sam's destiny, and that it's going to get him dead.

I confess, I am a bit of a nerd. I can't help myself :) I do still enjoy the show for the story and the handsome boys, but I enjoy it for all the clever extra layers as well. I hope my nerdy ramblings help to add something to the mix for you.
