No, you do not have to be familiar with all of CLAMP's works to join. So long as you know a few, you'll be fine.
As for how assignments are determined, the MODs assign people based on what they will write, what they will not write, so on. There is never a completely perfect match, since the requests are so broad in range, but so long as you specify the series that you will and will not write, then you'll be matched up with someone who wants a fic in the world of a series that you know.
