I'd like to qualify my "when I go to sleep" answer by saying that I have a walled-in yard, and a large dog...intruders aren't really that much of a threat for me.
I think my answer would differ if I didn't live in a house -- here I frequently leave the door open in the evenings (we have a screen door) if it's warm/cool enough depending on season. In college I used to base locking the door on whether or not I was going back out anytime soon - if not, I'd lock immediately, but if I was, I'd lock it on my way out.
My family lives off the street, has large dogs, and for the most part its a quiet neighborhood. Its just recently that we've started locking the normal lock at night when my parents get to bed, and the deadbolt gets deployed after the last person gets home.
But during the day, unless no one's home, we don't bother locking up.
