In the musical, just for clarity, she's "Miss Turnstiles," possibly because there really was a "Miss Subways?" And that was probably a bigger joke and made more sense when they put it in for the original audience. I had no idea that there really was such a thing!
I assumed everybody knew Miss Subways was real. I remember seeing the posters on the NYC subway when I was growing up. (Actually, they were ads that were above the seats, not big posters near where the doors were, but I don't know a specific word for that sort of advertisement.)
