Two things I have found to be consistent in the creation of humor and/or comedy. Conflict and Contrast.

Conflict happens all the time. Between children and parents, strangers on a train, bosses and employees. It's easy to twist one aspect of a conflict to make the conflict funny. Or to use the conflict as a springboard into a crazy situation.
