The cover is kind of funny. You've got Spider-Man and Wolverine, which you would expect from just about anything Marvel is putting out. Then you have Iron Man and Captain America, which makes sense since the game it about Civil War. Then you have Hulk, okay sure, if Hulk wants to be on a cover I'm not going to tell him differently. And Venom, huh? And then Deadpool, doesn't make sense but hey it's Deadpool, and Gambit, again huh? I can only guess DP and Gambit are there because they were in Wolverine Origins. And then, you can extend that out to all the characters (except Cap) as having been in a movie in the last few years. So yeah.

Hulk wasn't in CW at all, he was on Sakaar, boinking Caiera saving the planet and kicking asses. Gambit... wasn't even CLOSE that thing. Venom ♥

Iron Man being pro-reg and Cap being anti-reg are the only two affixed affiliations. You get to pick the side you're on and what characters you get to use. So the story's only loosely based on Civil War. The mutants are going to be more involved in this version, and the ones I can see on the official site are Wolverine, Gambit, Storm, and Jean Grey.
Oh yeah, other cool feature: unique two-character power combos. Thor can bounce his lightning off Cap's shield as they move and make an electric clothesline effect, among other nifty stuff.
