I'm wondering if the opening credits' "shattered glass" theme is a clue that Sam's soul is in fact in pieces, if the alphas hold those pieces and that's why the monsters are acting weird.

Is it possible that by the end of the season Castiel will want a human soul of his own? So he can feel again? Or that in some similar vein, Castiel who cares but isn't allowed to acknowledge it will parallel Sam who wants to care but can't?
Oh! That's a lovely theory and one I would love to see played out more.
You know what this makes me think of? Piers Anthony's Xanth series, where a half or even a quarter soul is enough to do the whole job. Share you souls pretty boys, share them! :D

I interpreted Cas' line to mean that he can't allow himself to feel. That he's repressing his emotions because he doesn't have the time to feel guilty over the things he has to do (this is my mind officially makes him a Winchester).
My brain likes to associate Castiel with Amalthea from The Last Unicorn by Peter S. Beagle. She was human too for a little while:
"I will go back to my forest too, but I do not know if I will live contentedly there, or anywhere. I have been mortal, and some part of me is mortal yet. I am full of tears and hunger and the fear of death, though I cannot weep, and I want nothing, and I cannot die. I am not like the others now, for no unicorn was ever born who could regret, but I do. I regret."
That, in my head, is Castiel.

I actually thought about this. And I have a way the show COULD resolve it, but it's much more like fanwank and probably has no chance of being canon. If I could fanwank my own explanations into canon, though, I'd say angels have Grace where humans have a soul. Because angels do have emotion indeed, but their vessels' bodies don't decay. So what if Cas had to choose between Grace and a human soul, which would absolve him of his responsibilities but would also make him subject to time's ravages? This would be scintillating to watch.
But then we'd need someone to run heaven in his stead.
