I'm still very behind in my Supernatural watching (last thing I saw was 5.08 Changing Channels). I've been re-watching Season 3 instead. How awesome was Ruby! This comment I made on a spnroundtable post about favourite female characters, pretty much explains why I liked her.
How do I choose? I love so many of the female characters, I even made icons of all the recurring ones. But I think that my favourite is probably Ruby. I spent all of Season 4 wondering exactly what she was doing. It wasn't until I saw this moment in On the Head of a Pin, with that little smirk I realised that she was manipulating Sam beautifully. Yeah, she was one of the bad guys, but she was so smart and made a fantastic villain.
