The pairing is actually Dean & a creature who kidnaps him, impregnates him, and throughout all of causes Dean to fall in love-stockholm style with it.
After he's given birth or been knocked up Sam finds them and kills the creature. I do remember that Dean gets really upset over it too.
If I remember right the creature isn't actually dead, just faked it.
