Maybe the first time he touched a living soul. I don't know; my current theory is that souls are the batteries that power heaven. The more of them up top or upstairs, the stronger the powers of Heaven. The more of them in purgatory, below, or corrupted, the weaker the powers of Heaven.
Yeah, that's probably it, but I wonder why having 50,000 more souls on earth would empower Cas but not Raphael? Assuming Cas wanted more soul-power to fight Raphael and not to fight hell or whatever.
