so apparently i'm not the idiot i thought i was. i've been having massive troubles with antibody staining now for about 3 months, perhaps a bit longer. i've followed protocols precisely, pHed all my solutions, repeated the experiment standing on only my left or right foot, sacrificed small mammals to the lab gods, etc. nothing seemed to be working. the only thing i haven't been changing was the secondary antibody i used. and no it didn't occur to me that the secondary might go bad til Lori mentioned it (mad props to Lori). behold, i too can make antibodies work (the data, i has it):


and as i said, antibody stains are pretty! so i'm happy to have one... hopefully it's the start of many more to come.
what started off as a super shitty week seems to be shaping up as a super awesome weekend. DDR with M-kun tomorrow night, a night out with the second years on Friday for Eileen's birthday, and then a DDR party on Saturday (and i'm not even the one throwing it this time). oh and like 5 mice to get through in the lab this weekend... so it will be a busy one. i'm starting the 3-D mapping project again on Monday, which is a good thing... i'm supposed to present a poster on it in 30 days.
i made a king cake for my lab (a shame my digital camera is dead) but no one was brave enough to try it at 9 am (too much sugar they said)... you people fail at being cajun (and that's probably a good thing...). it's strange but lately i've been homesick for Louisiana (don't ask, it makes no sense). also, Ekaterinn it was very nice hearing from you last night... your fandom truly amuses me... when are we meeting for cantarella and arsenic tea and cookies? i think it has almost been ten years now...
