In a list, because lists make things simpler:

1) It's pretty much canon that Rook holds his alcohol well. Considering that he's always on guard around Rolo, it's even less likely that he would get drunk to a point of being not in his "right mind"; there's no one in the world that he trusts enough to get shitfaced drunk in front of, to the point of being unable to handle himself.

2) At the end of Rook's last stay on Atia, Rook and Rolo had come to a tentative, if somewhat reluctant, ceasefire. Rook doesn't have a moral high ground in this situaion; given the opportunity, he would have revenge-killed Lelouch without a second thought, fully aware of what Lelouch means to Rolo, so on some level (albeit a very buried level), he understands Rolo's motivations, even though there are no forces in the universe powerful enough to compel him to actually forgive Rolo.

3) Rook is six inches taller than Rolo and at least 60 pounds heavier, if not more. Rolo's not stupid enough to try to pull anything on Rook and Rook knows it; he's beaten the shit out of Rolo in the past and they both know full well that if Rook really wanted Rolo to die, he could snap his neck one-handed.

4) With Havemercy off the island and the addition of two Airmen-one of whom is just as, if not more, attached to Thom as Rook himself is, and the other of whom Rook knows to be a very strong and capable fighter-Rook has far less of a reason to worry about Rolo's access to his weak points.

5) WRT Rook's original reply to Rolo's post, it was made almost entirely in jest. Rook likes to get a rise out of Rolo because he's just like that-Rolo's pretentious, holier-than-thou attitude annoys him, so he derives pleasure from goading Rolo to a visible reaction (which obviously worked, in this particular instance). As far as sex goes, he agreed because he's bored. He hasn't resumed work at Mao yet and, with the exception of his agreement to fight Ikkaku at some point in the near future, has no obligations to keep him occupied.

And on top of all that, sex with Rolo is, however primal it may sound, a way for Rook to exert dominance. Rolo is smarter than Rook is, more literate, better educated, and they both know it, which irks Rook to no end-so he takes the opportunities that he can to show ways in which he's superior. In this case, physically and sexually. With the single exception of Havemercy, Rook has never allowed himself to be dominated sexually-as a former prostitute, Rook is more than familiar with the use of sex as a tool of control and power, and Rolo is no exception to that attitude.

6) To tie points 4 and 5 together, since Rook has fewer weak points, he can more thoroughly humiliate Rolo in any way possible without fear of retribution. Hence a renewed willingness to engage with him sexually-sex with Rolo is nothing more than just a tool of dominance and humiliation, as I mentioned above. Rook doesn't equate sex with intimacy the way the majority of the island population does.

TL;DR - Rook's original comment was in jest, purely to get a rise out of Rolo, but the agreement was because unlike many people, Rook doesn't equate sex with intimacy or even fondness. He doesn't forgive and forget, and sex doesn't mean either-sex is a way for him to exert dominance, control, and humiliation over/onto Rolo.
