Kisuke's oldest and dearest friend, and the only person able to keep him in line... to some extent. He was raised with her in the Shihouin household and they've been inseparable ever since. Some time after they graduated from the Academy, their relationship evolved into a non-exclusive, romantic one. He served as her third seat when she took command of the Gotei 13's 2nd Division, and it was under her recommendation that he took the captaincy exams and was promoted to captain of the 12th Division. After he was framed by Aizen Sosuke for transforming the Vizard, it was Yoruichi who rescued him and their friend Tessai Tsukabishi and enabled them to escape Soul Society
Since their world was destroyed, their relationship has shifted yet again. Kisuke has become even more devoted to her, and spend most, if not all of his spare time with her to the point of practically taking up residence in her room. Thinking he had lost her opened his eyes to just how much he adores her, their previously casual, open relationship has become somewhat more exclusive. He's not complaining about it in the slightest.

Kisuke's student; Ichigo's progress as a Shinigami was closely monitored by him and when Ichigo needed to regain his powers after losing them in a fight to Kuchiki Byakuya, it was Kisuke's training which saw them restored to the teenager. Unfortunately this also resulted in the unfortunate side effect of Ichigo gaining a Hollow, but we don't talk about that. He also taught him how to fight properly instead of waving his vastly oversized zanpakutou around. He also is on good terms with Ichigo's Shinigami father Isshin.
Ichigo has only recently arrived on the Thor and Kisuke is delighted to see him; he can't help but feel somewhat responsible for him in lieu of his father's absence- not that he'll tell him that.

Once Yoruichi's most devoted subordinate, Soifon is certainly no fan of Kisuke's despite him not actually having anything against her. He finds her animosity towards him amusing and recognises it as being the product of jealousy of his closeness to Yoruichi.
Kisuke was relieved for Yoruichi's sake to see Soifon arrive on board the Thor and was shocked to see how badly she had faired in the battle against Aizen and his Arrancar in battle for Karakura.
