Why is it so difficult in immunohistochemistry to find the proper secondary antibody (with conjugate) that:
1. Has a signal that is detected at a level significantly higher than background;
2. Does not have nonspecific staining;
3. Has a peak excitation range for the lasers in your confocal microscope;
4. Doesn't cause fluorescence due to confounding factors (edge effects, autofluorescence, etc)
Why?
