Miss Marple. Although I'm not entirely sure if she actually qualifies as a detective, or if she was just a nosy old biddy.

Very few female detectives seem to be proper detectives. Holy Crap, I forgot Vickie!

Yeah, but she can fly. That's a bit of an advantage.

BTW: Veronica Mars & Nancy Drew may have been ommited because they are not licensed PIs. I'm not sure what Wikipedia's requirements are.

For amateur detectives I'm fond of Christie's Miss Jane Marple. Ditto Anthony Boucher's Sister Ursula.

Female teen amateur sleuths abound; Nancy Drew, Ginny Gordon and Trixie Belden come immediately to mind.

Does Mrs. Peel (from the Avengers) and Pat Savage (from the Doc Savage series)count as detectives? And how about amateur Egyptologist Amelia Peobody (from Elizabeth Peters)?

Miss Marple, as embodied by Joan Hickson in the 80s, is a definite favourite and like Jeremy Brett as Holmes, she nailed the role in such a way that I find it hard to watch anyone else as that character. The current ITV ones are absolute camp dross and mention of them is tantamount to swearing in our house.

I do enjoy Bones, if Temperance Brennan counts.
