Sam didn’t have many people in his life who believed in him. Not even Dean seemed to have that belief in him anymore. After the incident with Ruby and the demon blood, Dean couldn’t trust him. Which Sam thought was with good reason because, as he had admitted to Dean, he couldn’t trust himself either. So, they decided it was best for them to go their separate ways.
They never would have gotten back together if it hadn’t been for Zachariah sending Dean into the future. The year 2014, where Sam said yes to Lucifer taking over his body and the world was in ruins. Seeing the devil inside his brother’s body made it feel like his insides were being ripped to shreds. Or, to those who have no problems with “chick flick moments”, it felt like his was heart breaking. Lucifer told Dean he wouldn’t be able to change the future, that the world he saw would happen no matter what he did, but Dean wouldn’t believe it.
When he got back to his time, he called Sam and told him to come back. It wasn’t because he believed Sam would say no to Lucifer, it was the complete opposite. He believed that Sam would say yes unless he was with Dean, so Dean could stop it.
