I give enough tours of Durham, NC that it's amazing that I didn't think of doing this until now. Here's a (soon-to-be-updated) list of my favourite downtown spots and local resources - My particular faves (I tried to narrow it down) are marked with arrows:

DIRECTIONS / PARKING
From Raleigh-Durham International Airport: Take your best route to I-40 West.
Travel about 4 miles and use the right 2 lanes to take Exit 279B to NC Hwy 147 North. After about 8 miles, exit onto South Duke Street. When you reach Duke and Main Streets, you're downtown.
