I'd say that perhaps it wouldn't be a monogamous relationship on Dean's part, in the way that he'd sleep with other people, but then he'd want to be with Cas and maybe that wouldn't work either 'cause he'd be thinking about him.
I hadn't thought about this! It's an interesting aspect; I had told her, originally, that I could see him like not wanting to push Castiel into having sex with him because he's all freaking out over Cas not really being into physical intimacy and Dean is like all big damn hero and not wanting to do it because he's afraid it puts Castiel into an awkward position, so he masturbates like a beast and it's all frustrating because they're in a relationship (of sorts?) so Dean would feel guilty for picturing someone else, like another woman or man, but if he pictures Castiel it also feels awkward? Because for one they're in a relationship and for two it'd feel like a violation. Idek, I can just see Dean all ohmygod I can't take advantage of him and at the same time, he's a sexual being by nature so it's not like he can just turn the urges off. It'd be really interesting, to me, to see what kind of compromise they could come up with? Especially after delicious miscommunication and angsting and Castiel getting all frustrated because he isn't really opposed to having sex with Dean; being asexual doesn't mean he's against sex, it just means that's not really what he's about, that it's not a necessity for him. (That's not a generalization of asexuality as a whole; that's just a summary of Castiel's opinion.)

I agree with what you're saying. :) I mean the "violation" in a way that, as you stated, Dean would feel bad for thinking of Castiel when they're already in a relationship and he knows that Castiel isn't really into that and it just makes everything terribly awkward. (Dean is such a woobie, Jesus Christ.)
As for their sex life, I definitely can see Castiel wanting to do it because of a deeper emotional connection; I don't see he and Dean's relationship being based solely off of physical gratification. (Especially since this entire thing is about Castiel being asexual.) Furthermore, I agree with Dean being a sort of James Kirk a la Star Trek, needing that safety, stability, comfort and acceptance in a relationship. They both can and do have sex without strings attached; but when it comes to relationships, I don't see them as having just flings, or if they do, they're just that: flings. Dean would totally throw himself into a relationship when it came down to it, and with the chemistry between he and Castiel... well, it's not hard for me to buy that they're relationship would be quite devoted, if I may make such an assumption.
I can't really offer much on this because I haven't been a fan of Supernatural since mid-season 5, so my opinions on the characters could be entirely out-dated and Jossed. Either way, though, I'd still read this fic if she (or anyone, really) actually wrote it. I'm excited to see someone tackle this logically and not try to make it a "cure asexuality" fic, as is wont to do. (Not trying to insinuate that's at all what you are saying, though.)
