A second interview can be with different interviewers - e.g. this time with the actual reporting Line Manager and/or people from the team you'd be working with. Be sure to ask who will be interviewing you when the second interview is scheduled, and find out what you can about their positions, etc. If it *is* different people, you can ask some of the same questions: "As the reporting manager for this position, what do you feel are the most important qualities for the successful candidate to have?" etc. (And of course be sure to tell them you have all of those!) You could also ask them, since they're more likely to have closer knowledge of the post, what a typical work week would be in this position, what the key indicators of success would be, etc. To make a personal kind of connection with them, you could ask what *they* enjoy best about working at the company, or working with their team or work type.
Think over your first interview too - was there anything you feel you missed or should have said? Try to work it in there, perhaps by asking a question that allows you to expand a little based on *their* answer.
