Exactly the same as for traditional :) I draw digitally myself--it's even easier to set the page up than in real media. Just turn on the rulers, measure off the proper sizes with guides, then draw in the box boundaries for each area with blue lines--that'll be your template for the rest of your project. Draw on a separate layer, of course.

The results shouldn't vary depending on the order O___o What you should do is start opening a new file in Photoshop, and when the file spec dialogue window pops up, type in the print size you need (that's the page size in inches), and then the resolution per inch (600 dpi). Neither should change its value while you are typing the other.
