What is the significance of Same's blood trickling down?

I gathered that Sam had a mark or sigil of some kind on his skin that disguised him, and that by cutting it out with the knife he broke the spell. That was my take, anyway.

The blood is moving towards Dean, regardless of which way Sam holds his hand, so seems to me it would be a kind of 'blood calls to blood' thing. Blood affinity.

I imagined it as the blood flowing always towards Dean -- it's how Sam's tracked him across the post-apocalyptic world.
