I've been trying to lineage a cell for the past 7 months. It's definitely not supposed to take that long, but 4 of those months were kind of a bust due to TAing and almost another month was spent on vacation. Still, it's been a little ridiculous.
Today I finally met my cell(s, they're symmetrical on the left and right) of interest. Actually, I met them yesterday, but I have had too many hope-turned-disappointment moments to allow myself to be optimistic. But today I watched again, and the cellular landscape looked the same as yesterday.
The worm was great-- it moved around more than I would've liked, but it didn't flip over until the cell divisions were done and I had just taken some decent pictures. So then I took pictures of the cell on the other side, and then after that it flipped back over so I could take some later pictures on the first side, and so on.
I've been waiting for this for so long.
So what did I do now that I met my cells? Tried to kill them. Hahah... so evil... but actually, that is one of the next experiments that needs to be done, and it's one that can be done now without waiting to watch anything else. In reality I probably didn't succeed in killing them-- I've always been bad at video games. The next step is to time it so that these cells are born in the morning instead of 9 pm, so that I can actually watch them migrate around and then die four hours later.
I also submitted an abstract for the poster session for this summer's worm meeting. So all in all today has felt pretty accomplished.
On the way home I could smell the mint-chocolate factory. It was not too cold tonight. Friday's temperatures got up to 21 C/70 F, and then it snowed yesterday. It didn't stick though-- I think it's almost spring.
