Well, if you're not picky about genre, here are some of my favorite series at the moment:
Kitty and the Midnight Hour by Carrie Vaughn
Kitty is a werewolf. And an awesome dj with a program called The Midnight Hour, which is basically Loveline for the supernatural. Personally, I like this series because of the pack dynamics--it starts off with the standard aggressive-alpha-male-dominates-young-new-female-wolf dealio, but quickly moves into Kitty carving her own place for herself. I also really like this series because it separates the human and wolf sides a little more than some series. When Kitty is a wolf, she's a wolf, and when she's human, she's pretty human. None of the the-alpha-howls-and-everyone-is-magically-spellbound stuff.
Stardoc by S. L. Viehl
Set about 2500 years in the future, iirc, a young, brilliant, surgeon named Cherijo abandons earth and sets off for an alien colony to become a trauma surgeon with nothing more than her fierce surgical skills and her iron will to guide her. She's running from her father, who did something AWFUL and MYSTERIOUS (there's plenty of backstory as the series goes on), and basically, runs into a bunch of allies and enemies and has to forget everything she knew about medicine and learn how to treat aliens she's never even SEEN let alone studied before.
Nightlife by Rob Thurman
Okay, disclaimer, I'm only halfway through the first book at the moment, but it is some of the best urban fantasy I have ever read, so. If you like truly dark urban fantasy, with monsters who are actually monstrous, and spunky, smartalecky young male protagonists, who always, always open their mouth when they should just shut it, and brothers who will go to hell and back for each other, then you should totally check out this book. And now that I think about it, you probably should START with this book, because you like Supernatural, and yeah.
Boneshaker by Cherie Priest
Another book that I haven't quite finished, and not part of a series, but just throwing it out there, in case this is your thing. It has airships, zombies, steampunk, alternative history, kick-ass mama bears, etc.
And then Kelley Armstrong's Women of the Underworld Series
Personally, I've only read about half of the third book (which I totally thought was the first book, or I would've started with book number one), but it's basically an entire world of supernatural species--witches and sorcerers and werewolves and stuff--and there's multiple major characters that the various books focus on from what I can tell, but they're all awesome, strong, kickass, realistic female characters. If you're jonesing for some estrogen.
