Well, I thought about it for a bit...I'm not sure if I'm the best person to give suggestions, but I know there's worse out there. Some of it is personal extrapolation, some of it firm BS, some of it canon.

Itsuki is by far the most mysterious of the SOS Brigade, with the least amount of backstory fleshed out. This is both a strength (allows the player to make up details as they go along) and a weakness (there's not as much of a framework as other characters to go on) but what we know is this: there's a before and an after in his life, and the divide comes from that moment in which he discovers that he's an esper.

He wasn't born poised and smarmy, and it's pretty obvious that it's a smooth plastic outer shell with his true self different from the facade he presents to the SOS Brigade. He was different when he was younger, and when that transition was is...hard to say. (I vaguely remember reading a quote in which he alludes to looking for aliens and ghosts and mysteries when a child, although it might've just been my bad memory.)

It's hard to say exactly how much of what he does is an act and how much of it is actually him. Things I think are safe to assume: he enjoys mysteries, puzzles, riddles, and enigmas. He enjoys being a part of the SOS Brigade and being caught up in their misadventures, though he can take or leave the giant fighting part. (Or at least tone it down in frequency.) One quote from the light novels...ah, found it:

"That's okay, isn't it? This is enjoyable work. An activity to protect a small and weak club; high school students making a resistance against the big and powerful Student Council......."

Koizumi had become rather eerie as his refreshing eyes got this distant look in them, before he regained his smile.

"The truth is, I have fantasized about such a school-life. More and more, I am acknowledging Suzumiya-san's divinity, and at times I've felt like I wanted to kneel down and worship her. Because she has granted me my dream."

Sexual orientation issues aside, Itsuki is genuinely interested in Haruhi. In the alternate timeline in The Disappearance of Suzumiya Haruhi he's her classmate once more, and claims that he's interested in her...if he's telling the truth or not you can decide, but point being in a canon AU he's still interested in her.

She's a puzzle both in how she gained her God powers, how she'll end up expressing her God powers, and just generally as a person. Gay, straight, bi, or asexual, Haruhi is always going to be one of Itsuki's first priorities...not only because of her God-powers but I think also because she's an interesting person through which Itsuki was able to live a lot of his fantasies he otherwise gave up on.

How he'd interact with her depends on how much she knows, but I think it's safe to say Itsuki wouldn't treat her like an idiot but not go out of his way to tell her more than she needs to know, and if he says no to her it would only be for an extraordinarily good life-or-death reason. Because issues aside, they've got roles and his is to be the perpetually smiling yes man.
