Well, I'll have a go. The scene in Mystery Spot, when Sam pulls Dean back from getting run over by the old man. Dean's all keen to find out what it looked like, and a slighly pissed Sam tells him, 'you peed yourself'. I just love Jensen's disgusted face and subsequent defense.

I know. Cerebral over slapstick, but it was the only scene that made me laugh out loud (in the entire series actually). Thus, it is my favourite funny scene.
