Transformers G1, any canonically female Cybertronian/any canonically female Cybertronian, gender seemed like such a nebulous, immaterial thing--until she stared a relationship with another femme.

Buffy: the Vampire Slayer, Dawn Summers, Willow made being out look so easy, but for Dawn, things weren't nearly so simple; she was supposed to be the normal one, after all.

Some of these might be repeats from previous lgbtfest prompts.

Star Trek: DS9, any Joined Trill, Being Joined has been known to influence the host's orientation or gender identity.

Star Trek: DS9, Ezri Dax, Ezri's family was not in favor of her being Joined, and they definitely aren't in favor of some of the (gender or sexual orientation) changes.
