Awesome list! I've got some friends who are huge fans of Buffy but haven't gotten the chance to watch Supernatural (though they want to...especially since I've gone on and on about it, lol...plus I've been insisting that Dean is perfect for Buffy whenever the topic comes up ^_^) and I'll definitely point them in this direction.
Oh, I also thought of something for these two. Children. They've both had to tell little kids what's really out there in order to try and protect them.
Dean: I'd give anything not to tell you this. But sometimes nightmares are real. Micheal: Then why are you telling me? Dean: Because we need your help. Micheal: My help? Dean: We can kill it. Me and him. It's what we do.
Buffy: Grown-ups don't believe you, right? Well, I do. We both know there are real monsters. But there are also real heroes that fight monsters. And that's me.
Guy: How'd you do that? Buffy: It's what I do. (Okay, so this last one wasn't to a little kid but it mirrored what Dean said, so I used it.)
And I also thought of another quote for #9. (I know, this is going to end up being a long comment but once I started thinking on this, I couldn't stop! LOL)
Faith: See, you need me to toe the line because you're afraid you'll go over it, aren't you, B? You can't handle watching me living my own way and having a blast, because it tempts you. You know it could be you.
Buffy (talking to Willow about Faith going bad): I know Faith isn't exactly on the cover of Sanity Fair, but she's had it rough. Different circumstances, that could be me.
And a whole new one to add to your list would be the way they look at their job. More specifically, how they see their mentors. In season one, there's this exchange:
Sam: I just don't see how you can have so much faith in him, how you can just blindly follow orders. Dean: Yeah, it's called being a good son.
and
Dean: I didn't follow Dad's orders and it almost got you killed.
In season three, Buffy and Faith have this exchange:
Faith: You actually gonna take orders from him? Buffy: That's the job. What else can we do?
Then later, as the series/season goes on they both begin to rebel in their own ways. Buffy quits the Watcher's Council (but still mostly does what Giles tells her because she still considers him her Watcher). Dean finally goes off on his dad when John is trying to get pissed at him for not calling about Sam's visions and tells him, "I called you when we were in Lawrence. Sam called you when I was dying. Call you? I got a better chance at winning the lottery than having you pick up." and he also uses a bullet to save Sam, knowing that his dad would be pissed, which he realizes later when he figures out John's possessed. "He wouldn't be proud of me. He'd be pissed that I wasted a bullet."
I'm pretty sure I had quotes to support #6 as well, but this is already way long...so if I do, I'll just comment again. *g*

Thank you for all the quotes! I put some of them in and you inspired another reason.
For some reasons, it is hard to find a quote, much less a really good one. Others there are tons of quotes. It can be hard to decide on the best one.
Plus, BtVS went for 7 seasons so there is much more information and dialouge than there is for SPN. It can be hard to find a quote for Dean sometimes b/c there just haven't been enough episodes.
Like we KNOW that being a hunter is WHO Dean is. We don't need a quote to tell us that and until very recently, there wasn't one.
