I've had many many people ask me about how to do animations in icons. Most of my knowledge comes from the lovely mannequinskin, but I've taught myself a few things as well. This tutorial expands to AS3 as well, so you won't be able to do this without an animating program of some kind.

Lovely tutorial. I used it to do this in PS8 and Image Ready because there are less steps you need to follow.
