Well, according to Romanian legend, being blinded then buried alive (brutal murder) is enough to turn one into a vampire o.o Also, unfinished business can result in a dead person rising as a vampire. This could be enough reason for Vlad and Mircea; Vlad never really punted the Turks all the way out of Romania, Mircea could rise for revenge. Radu, however, is more of a mystery to me. He died of syphilis; he wasn't murdered. :o So I guess, like you said, he was born into it, but only became one after he died. :3 Also, this makes me wonder; When Vlad found out Radu had risen, did he maul the hell out of him? I would have. >:[ Or at least smacked him a good one.
However, with the Mircea and the buried alive thing, according to some source of which I can't remember the name of (I think it might be VAMPIRE The Complete Guide to the World of the Undead by Manuela Dunn Mascetti, but I'm not sure), Mircea was buried face down, so he wouldn't rise as a vampire. D: As in, he'd dig in the wrong direction. Journey to the Center of the Earth, anyone? XD Also, this might be why it must have been later for him; I mean, after a while, he's going to realize he's goin' the wrong direction and be like, "Aw SHIT!" and go the other way...Although, he must have been digging for a really long while if it took him THAT long.
But, I can't be sure, because the above mentioned book is so full of crap that it makes me giggle. That and it's extraordinary ability to make normal graveyard pictures seem as if they're just horribly evil places where rabid vampire rats come out to suk yur bluuuuud.
Also, I don't actually believe they're actually vampires, it's all hypothetical and I'm not crazy I swear. :
